Mitochondrial haplogroup L3 formed within Africa and later became the key maternal branch ancestral to almost all non-African mtDNA through its descendants M and N. Soon after L3 formed, two major daughter branches—macrohaplogroups M and N—diversified and became the foundation of most maternal lineages outside Africa. Reviews of the human mitochondrial molecular clock emphasize that different calibrations and the effects of selection can shift estimated dates, which is why many timelines use date ranges rather than single numbers.
One route model argues the main successful dispersal passed through northeast Africa into the Levant. Whole-genome comparisons of Egyptians, Ethiopians, and non-Africans have been used to support Egypt as a likely gateway, showing Northeast African haplotypes closely related to non-African variation after masking later West Eurasian ancestry.
As mtDNA sequencing expanded, researchers maintained curated phylogenies to keep haplogroup definitions consistent. PhyloTree (commonly used through tools such as HaploGrep) provides a widely used, regularly updated tree that helps ensure that “L3,” “M,” and “N” mean the same set of mutations across studies.

The outcome is a clearer, testable framework: most living non-African maternal lineages trace back to L3 via M and N, while continuing debate focuses on where and how that spread occurred.
